I've recently bought a new desktop machine which cames with Windows Vista. I wasn't particularly eager for Vista; DRM and COM for the new driver model, among other things, wasn't very appealing. After using it for a couple of weeks, it's OK (I admit I liked XP a lot). For developers interested in multimedia, the fact that Vista requires DirectX 9 creates a new baseline with good features. Vista comes with the .Net 3.0 runtime, and its presentation framework, WPF, I haven't looked into this much yet. But I've started playing with Silverlight, and I found this quite interesting. Its version 2.0 will allow scripting with .Net languages. If you want to get started with minimal fuss, David Miles has a couple of interesting tutorials, for example, here.
